ADH1B (alcohol dehydrogenase 1B) is a key gene in alcohol metabolism. The rs1229984 variant (ADH1B*2) encodes a fast-metabolizing enzyme that rapidly converts ethanol to acetaldehyde, causing unpleasant effects like flushing and nausea. This often leads to lower alcohol intake, indirectly reducing risks of alcohol-related diseases (liver cirrhosis, certain cancers). Some observational and GWAS studies link this variant to increased longevity, but the evidence is moderate: it is based on association studies, not RCTs. The effect is indirect and mediated by behavior. Caveat: the variant does not protect against harm from heavy drinking, and individual outcomes depend on many genetic and lifestyle factors.
