The vitamin D receptor (VDR) is often discussed in the context of longevity because vitamin D influences many age-related processes (immune function, bone metabolism, cell division). The rs2228570 (FokI) variant in the VDR gene slightly alters receptor function. About 38% of Europeans carry the A allele. However, the effect on cellular vitamin D response is small; blood vitamin D levels are far more dependent on sun exposure and diet than on genetics. There is no robust evidence from randomized trials directly linking VDR variants to extended lifespan. Observational studies show associations between low vitamin D levels and increased mortality, but causality is not proven. Genotype-based supplementation is not currently recommended. Caution is warranted: high-dose vitamin D without medical need can be toxic. In summary, the evidence for VDR and longevity is weak to moderate – mainly from GWAS and mechanistic studies. Lifestyle factors (exercise, diet, sunlight) have a much greater impact on healthy lifespan.
