Is TCF7L2 useful for fatigue?

TCF7L2 is one of the most robustly replicated genetic risk loci for type 2 diabetes, identified through large GWAS. Variants like rs7903146 increase diabetes risk by affecting pancreatic beta-cell function. However, there is no direct evidence linking TCF7L2 to fatigue. Fatigue is a multifactorial symptom; while undiagnosed diabetes can cause tiredness, the gene variant alone does not predict current energy levels or fatigue. Some direct-to-consumer genetic tests market TCF7L2 as a 'fatigue gene' or 'energy gene', but this is not supported by clinical evidence. The American Diabetes Association and other guidelines do not recommend TCF7L2 testing for fatigue or general health.
