How are TCF7L2 and longevity related?

TCF7L2 is a transcription factor central to Wnt signaling. Multiple GWAS have robustly linked variants (e.g., rs7903146) to increased type 2 diabetes risk. Since diabetes is a major age-related disease, TCF7L2 may indirectly affect longevity through metabolic complications. However, direct evidence for an association with lifespan or healthspan is weak.
