Can VDR help with Darmgesundheit?

The vitamin D receptor (VDR) is a nuclear receptor that mediates the effects of vitamin D. In gut health, VDR plays a plausible role: it regulates tight junction proteins (e.g., claudin, occludin) and thus may influence intestinal barrier integrity. VDR also modulates innate immunity and gut microbiota composition. However, the evidence for a direct, clinically actionable effect of VDR gene variants on gut health is moderate. Most data come from cell and animal models or observational studies with small effect sizes. A consumer DNA test reporting VDR variants cannot diagnose or guide treatment for 'leaky gut' or other gut disorders. Gut permeability is multifactorial (diet, stress, medications, microbiome), and a single genetic marker is rarely decisive. A more useful approach includes blood biomarkers (vitamin D, calprotectin) and medical evaluation. VDR is an interesting piece of the puzzle, not a magic bullet.
