MC4R (Melanocortin-4 Receptor) is a key regulator of appetite and energy homeostasis. There is evidence that MC4R also plays a role in the stress response, particularly in stress-induced eating behavior. Animal models show that stress can alter MC4R signaling in the hypothalamus, leading to changes in feeding. In humans, certain MC4R variants are associated with higher BMI and emotional eating, but the direct link to stress mechanisms is not yet firmly established. Most studies are associative or mechanistic; large prospective human trials confirming a causal role of MC4R in stress responses are lacking. Moreover, consumer DNA tests typically detect common variants with small effect sizes. Clinical relevance for stress management is therefore currently limited. Evidence: mechanistic/moderate. Caveat: single nucleotide polymorphisms often have small effects; blood or saliva tests for stress markers are more actionable.
