How are FOXO3 and allergy related?

The relationship between FOXO3 and allergies is not yet firmly established. FOXO3 is a transcription factor involved in aging, oxidative stress, and inflammation. Since allergies involve immune dysregulation, FOXO3 variants may modulate allergic inflammation. Some GWAS have reported associations between FOXO3 SNPs and asthma or elevated IgE, but effect sizes are small and replication inconsistent. Evidence is therefore mechanistic or GWAS-based, not clinically robust. Consumer DNA tests that claim to predict allergy risk from FOXO3 variants overstate the science. Currently, no actionable recommendations for allergy prevention or treatment can be derived from FOXO3 genotyping. Established factors like allergen exposure, microbiome composition, and lifestyle remain far more relevant.
